Abstract
Among a class of novel ester conjugates, propofol linoleate has emerged as a potential antineoplastic agent. In the present study, the antigenotoxic and the antioxidative efficacy of propofol linoleate was explored against doxorubicin (DXR)-induced toxicity using cytogenetic, biochemical as well as histological analyses in murine model. Significant increase in chromosomal aberrations, micronucleated polychromatic erythrocytes (MNPCEs), lipid peroxidation and decrease in antioxidant enzymes was observed in animals of DXR treated group. Histological observations also revealed that DXR caused marked changes in hepatic and renal tissues of mice. The pre-treatment with propofol linoleate showed attenuation of cytotoxic damage and histological changes induced by DXR. The conjugate also acted as antioxidant, reducing the oxidative stress by elevating the activity of antioxidant enzymes. The important protective mechanisms offered by the conjugate from DXR-induced genotoxicity might be free radical scavenging property, DNA binding potential or increase in the antioxidant status.
