Abstract
•Three methods were developed and validated for the determination of valganciclovir hydrochloride (VLGH) in pharmaceutical formulations.•Two titrimetric methods were based on the titration of VLGH in acetic acid with HClO4 in the presence of Hg(CH3COO)2.•One spectrophotometric method was based on the reaction of VLGH with DMAB and the resulting yellow color was measured at 420 nm.•The paper describes the first report on the use of titrimetric technique for the determination of VLGH.•The three developed methods are simple, rapid and successfully applied for the determination of VLGH drug in pure form and in tablets.
Two non-aqueous titrimetric and one visible spectrophotometric methods were developed and validated for the determination of valganciclovir hydrochloride (VLGH) in pure drug and tablets. The titrimetric methods were carried out by titrating the VLGH solution in glacial acetic acid with acetous perchloric acid (HClO4) in the presence of mercuric acetate and the end point of this titration was being located either visually (method A) or potentiometrically (method B). The spectrophotometric method (method C) was based on the addition of p-dimethylaminobenzaldehyde (DMAB) solution to VLGH solution and the formed yellow condensation product was measured at 420nm. In titrimetric procedures, both the methods A and B were applicable over the range of 4–20mg VLGH, and the calculations were based on a 1:1 reaction stoichiometry (VLGH:HClO4). In a spectrophotometric method, Beer's law was valid in a concentration range of 5–50 μg/mL VLGH with the corresponding value of molar absorptivity of 4.43×103L/mol cm for method C. The limits of detection (LOD) and quantification (LOQ) for method C were 0.3 and 0.92μg/mL, respectively. The developed methods were successfully applied to the determination of VLGH in tablets, and the results were statistically compared with those of a reference method by applying Student's t-test and F-test. Further, the validity of the developed methods was confirmed by recovery studies via standard addition technique.
Deux méthodes titrimétriques non aqueuses et une spectrophotométrique visible ont été développées et validées pour la détermination du chlorhydrate de valganciclovir (VLGH) dans le médicament pur et les comprimés. Les méthodes titrimétriques ont été réalisées en titrant la solution de VLGH dans l’acide acétique glacial avec de l’acide perchlorique acéteux (HClO4) en présence d’acétate mercurique et le point final de ce titrage était localisé soit visuellement (méthode A) soit potentiométriquement (méthode B). La méthode spectrophotométrique (méthode C) était basée sur l’addition d’une solution de p-diméthylaminobenzaldéhyde (DMAB) à la solution de VLGH et le produit de condensation jaune formé a été mesuré à 420nm. Dans les procédures titrimétriques, les méthodes A et B étaient applicables sur la plage de 4–20mg de VLGH, et les calculs étaient basés sur une stoechiométrie de réaction 1:1 (VLGH:HClO4). Dans une méthode spectrophotométrique, la loi de Beer était valide dans une plage de concentration de 5–50μg/mL de VLGH avec la valeur correspondante de l’absorbance molaire de 4,43×103 L/mol cm pour la méthode C. Les limites de détection (LOD) et la quantification (LOQ) pour la méthode C étaient de 0,3 et 0,92μg/mL, respectivement. Les méthodes développées ont été appliquées avec succès à la détermination de la VLGH dans les comprimés, et les résultats ont été statistiquement comparés à ceux d’une méthode de référence en appliquant les tests t et F de Student. En outre, la validité des méthodes développées a été confirmée par des études de récupération via une technique d’addition standard.
