Abstract
Immunoflourescent and histopathological studies, gross lesions and clinical signs were observed in experimentally Newcastle disease virus (NDV) infected broiler chicks. From 4(th) day post inoculation (DPI), chicks were anorexic, suffered from greenish diarrhea showed respiratory signs with nasal discharge and difficult breathing. Torticulus and in-coordination was also seen during this period. Tracheal hemorrhages and splenomegaly was seen after 4(th) DPI. Alveoli of lungs were filled with edematous fluid and infiltrated with mononuclear cells. Lymphocytic degeneration in bursa and lymphoid hyperplasia in spleen were recorded on 8(th) and 10(th) DPI. Hyper immune serum against NDV was raised in rabbits, purified and globulins were conjugated with fluorescence iso-thiocyanate for immunoflourescent staining of slides for the detection of virus. Slides were deparaffinized, stained with FITC-conjugated antibodies to see under fluorescent microscope. Immunofluorescence was seen in trachea, lungs, spleen and bursa from 4(th) at various rats characterized as low, moderate and high. (C) 2011 Friends Science Publishers
