Abstract
An intracellular lipase was isolated from Aspergillus niger and the enzyme was purified by gel filtration chromatography on Sephadex G 75 column. The purified enzyme was found to be homogeneous as judged by electrophoresis on polyacryl amide gels. The specificity of the lipase towards single fatty acid triacylglycerols and natural triglycerides was determined. The enzyme preferentially attacks tripalmitoylglycerol as compared to other triglycerides. Positional specificity on using milk fat and olive oil triglycerides showed that the enzyme attacks positions 1 and 3 in triglyceride molecules.
