Abstract
A protocol for continuous high frequency regeneration of Vanda teres through long term-culture of nodal segment derived protocorms was achieved in Vacin and Went (VW) medium. Nodal segments cultured on VW medium with 2.0 mg l(-1) Kn + 0.5 mg l(-1) NAA + 2 % sucrose + 15% coconut water (CW) + 2 g l(-1) peptone produced an average of 25 protocorm like bodies (PLBs) within 4 months. PLBs were multiplied by subculturing on the same nutrient medium, where a PLB multiplied into 10-15 PLBs within 2 months. Shoots from PLBs were achieved by subculturing the PLBs on VW medium supplemented with 2% sucrose + 2 g l(-1) peptone + 1 g l(-1) active charcoal + 15% CW + 150 mg l(-1) L-glutamine within 2 months and by this time new PLBs induced from the base of the developing shoots. Shoots rooted on VW medium supplemented with 2% sucrose, 2 g l(-1) peptone, 15% CW, 1 g l(-1) active charcoal, 1.0 mg l(-1) IBA with or without 50 g l(-1) banana pulp. By repeated subculture of PLBs on plantlet regeneration medium, plantlets as well as PLBs were produced continuously. About 8 months were required from PLBs induction to the production of plantlets for transplantation.
