Abstract
The preparation and development of primary reagents of thyroglobulin (Tg) radioimmunoassay technique with low cost is considered to be the main objective of present study. The production of polyclonal antibodies of thyroglobulin was undertaken by immunizing three bulb/ C mice intraperitonial through primary injection and two booster doses. The preparation of I-125-Tg radiotracer was carried out using chloramine-T as oxidizing agent. The preparation of thyroglobulin standards were carried out. Optimization and validation of the assay were studied out. The results obtained provide a highly sensitive, specific and accurate RIA system for thyroglobulin based on liquid phase separation. In conclusion, this assay could be used for diagnosis of thyroid cancer.
