Abstract
Ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS-MS) method which is simple, sensitive and rapid was developed for quantitative determination of gefitinib in human plasma. This methadology is a excellent analytical approach for rapid analysis without compromising the efficacy. Protein precipitation technique with acetonitrile was utilized for preparation of samples and the chromatography was done on C-18 Acquity UPLC BEHTM column with neratinib as internal standard (IS). The mobile phase used for the chromatography was acetonitrile/water (70:30, v/v) and each of them contained 0.1% ammonia. The analysis was performed in ESI+ mode m/z 447.120 -> 128.0 for gefitinib and m/z 557.51 -> 112.17 for the neratinib (internal standard). The linearity of the method was established between 0.7-200 ng/mL and the method developed being simple, fast and extremely sensitive, could be used for quantification of gefitinib in human plasma.
