Abstract
The title compounds were prepared by cyclizing salicyldehyde with diethylmalonate in presence of piperidine to get 3-carbethoxycoumain that was reacted with hydrazine hydrate in ethanol to obtain 3-carbohydrazidocoumarin. The condensation of 3-carbohydrazidocoumarin with isatin followed by reaction with formaldehyde and different aromatic amines resulted in title compounds. All the synthesized compounds were characterized on the basis of their IR, H-1 NMR, mass and elemental analysis. The anticonvulsant activity of title compounds revealed that four compounds (Ar=4-bromophenyl, Ar=2-nitrophenyl, Ar=4-fluoro-3-chlorophenyl and Ar=2-methylphenyl) exhibited highly significant and comparable anticonvulsant activity with respect to standard drug phenytoin. The structure activity relationship indicated decrease in activity by replacing phenyl ring with triazole moiety. Most of the compounds possessing electronegative groups at C-2 and C-4 positions displayed good anticonvulsant activity. An electron releasing group (CH3) at C-4 also exhibited significant activity.
